Subsequently, we conducted the co-culture of HER2-positive and HER2-negative cell lines at varying ratios,[52] followed by treatment with the abovementioned ADCs. The viability of HER2-negative cells in the co-culture system was measured by detecting the GFP fluorescence intensity. The results showed that three ADCs with exatecan derivatives payload killed both NCI-N87 and MDA-MB-231/GFP cells, but T-DM1 (negative control) did not, which was consistent with the result observed by fluorescence microscopy
