Antibody Function H231 inhibited EREG from hEREG-293T cells.
Antibody Antigen Binding Assay 293T cells stably overexpressing hEREG, mEREG, or vector or colorectal cancer cells were seeded onto poly-D-lysine-coated black 96-well plates and incubated overnight. Serial dilutions of H231, cmAb, H231 ADCs, or cADC were added for 2 hours at 4°C. Plates were washed in PBS, fixed in 4% formalin, and incubated with anti-human Alexa-555 (Thermo Fisher Scientific, cat. #A-21433, RRID: AB_2535854) for 1 hour at room temperature. Fluorescence intensity was quantified using a Tecan Infinite M1000 plate reader (RRID: SCR_025732). Each condition was tested in triplicate in 2 to 3 independent experiments.
