By Cathepsin BDrug release by Cathepsin B was evaluated in vitro by incubating the ADC (1.5 ug/mL) in the presence of human Cathepsin B (Calbiochem #219362, 40 ug/mL) in acetate buffer pH 5 at 37°C. for 24 h and detecting the drug by UPLC-MS-SIR-DAD-ELSD. The slope was determined based on the kinetics over the 1st hour, the half-life on the 24 h kinetics.
